Antibody Humanization
Antibody humanization is the process of substituting the framework region and complementary determining region (CDR) of an antibody obtained from a non-human species to increase their similarity to the antibodies found naturally in humans. Antibodies derived from non-human species should not cause any adverse immune responses in the body in order to be developed as a therapeutic drug. In the process of humanizing the antibody, it is necessary to modify the different protein sequences to enhance their similarity to the antibodies in the human body, while maintaining its ability to recognize the target antigen at the same time.

Shuffling
The CDR of an antibody is directly involved in binding to an antigen, while the framework region is associated with the structural characteristics. The ability of an antibody to bind to an antigen is determined by the specificity determining residue (SDR) of the CDR. SDRs are usually located in CDR2 and CDR3 of the VH and CDR1 and CDR3 of the VL.
YBiologics uses its proprietary fully human antibody library Ymax®-ABL, and frame-region shuffling method, a proprietary technology to incorporate the CDR for the production of humanized antibodies with an optimized combination between mouse CDRs and human framework regions.

- Only the CDR3 of the variable region of the heavy chain is maintained as the CDR3 region of the mouse antibody
- Human antibody base sequences are used for the rest of the variable region of the heavy chain and the variable region of the light chain using human antibody nucleotide sequences
